EBOV minigenome assay. 293 cells were transfected with siRNA against the nontarget or Sec61α. Forty-eight hours later, the cells were transfected with various amounts of plasmid for the expression of VP24 or influenza virus M1 (M1), along with plasmids for the expression of NP, VP35, VP30, L, the T7 polymerase, Renilla luciferase, and an EBOV minigenome possessing the firefly luciferase gene. At 48 hours after transfection, the cells were lysed and the luciferase activity was measured.
