Overexpression of TID-containing fragments of Atg1 led to a disruption of the Atg1 self-interaction. A, overexpression of the 331-C fragment of Atg1 resulted in a diminished level of the Atg1-Atg1 complex. The 1–330 and 331-C fragments of Atg1 were overexpressed in cells that were also expressing HA- and myc-tagged versions of the full-length Atg1. The Atg1 self-interaction was detected by Western blotting for the presence of the Atg1-myc protein in immunoprecipitates that had been collected on an α-HA affinity resin.
