Caspase-8-mediated cleavage of IRF-3 is an intermediate step in its proteasome-mediated degradation. IRF-3 undergoes proteasome-mediated degradation in response to dsRNA-dependent signaling. Stimulation of TLR3 or RIG-I signaling by dsRNA activates caspase-8 by proteolysis of inactive procaspase-8. The activated caspase-8 initiates proteolytic processing of IRF-3, generating an intermediate product. The cleaved IRF-3 is then targeted for poly-ubiquitination en route to further degradation.
