Degrons function at the C terminus of the substrate polypeptide. Constructs expressing eGFP fusion proteins containing degrons at their C termini were stably transfected into cell line RJK88.13 and treated with CHX. Decay of eGFP concentrations was monitored over the indicated times by Western blotting (see “Experimental Procedures” for details).
