LIMK1 knockdown decreases spin-mediated enhancement. (A) The LIMK1 knockdown cell line, shLIMK-007, and the control cell line, shNTC, were examined by Western blotting using an anti-LIMK1/2 antibody or an anti-glyceraldehyde-3-phosphate dehydrogenase (GAPDH) antibody for the loading control. (B) These cells were also examined by F-actin staining with FITC-phalloidin. (C) shLIMK-007 or shNTC cells (2 × 105) were infected with HIV-1NL4-3 (200 ng p24) or spinoculated (at 300 × g or 1,200 × g) for 2 h, washed, and then incubated for 4 days. Viral replication was monitored by p24 release. The fold enhancement of viral replication by spinoculation was calculated using not-spun infected cells as “1.”
