A, RANK- and TAB2-induced NF-κB activation is blocked by TAK1-C100. HEK293 cells were transiently transfected with the indicated constructs together with an NF-κB luciferase reporter vector. After 36 h, cells were harvested and luciferase activity was measured as described in “Experimental Procedures”. Expression of the indicated proteins was detected by immunoblotting the lysates with anti-FLAG (lower panel). B, RANK, TRAF6, and TAB2-induced NF-κB activation is dependent on the interaction between TAB2/3 and TAK1. HEK293 cells stably expressing GFP or TAK1-C100 were transfected with the indicated constructs together with an NF-κB luciferase reporter vector. Cells were processed as described in (A).
