DBA mice were immunized i.d. with cII/CFA and spleens were collected two weeks later. Single cell suspensions were re-stimulated in vitro with 50µg/mL heat-denatured collagen in the presence of recombinant cytokines (A) or 10µg/mL purified cytokine-neutralizing antibodies (B) for five days. Supernatants were collected and IL-17 was measured by ELISA. Error bars represent SEM of triplicate culture samples. *p<0.05, **p<0.01, ***p<0.001 versus 0ng/mL (A) or versus isotype control (B) by one-way ANOVA. (C) BALB/c mice were immunized i.p. with KLH in CFA and spleens were collected two weeks later. Single cell suspensions were re-stimulated in vitro with 50µg/mL heat-denatured KLH in the presence of 10ng/mL recombinant cytokines or 10µg/mL purified cytokine-neutralizing antibodies for five days.
