Effect of fatty acid chain length and saturation on percentage of substrate hydrolyzed by PNPLA3

TAGs and DAGs containing a single species of fatty acid (myristate, palmitate, oleate, linoleate, and arachidonate) were prepared as substrates at a concentration of 200 μm. Substrates were incubated with purified PNPLA3 at 37 °C. The acylglycerol substrates (TAG or DAG) and released free fatty acids were isolated and quantified by GC-MS. The percentages of hydrolyzed substrates were calculated. Each reaction was repeated at least three times. NA, not applicable.
