Immunodetection of 4HPPD in subcellular fractions of green cultured cells of Arabidopsis. Lane 1, Purified protoplasts (100 μg); lane 2, enriched chloroplastic fraction (100 μg); and lane 3, enriched cytosolic fraction (100 μg). Total extracts were separated on a linear gradient of 3% to 27% (w/v) acrylamide in nondenaturing conditions and analyzed by western blotting using a 1:2000 dilution of an immunopurified polyclonal rabbit anti-carrot 4HPPD antibodies (Garcia et al., 1997) revealed by chemiluminescence.
