The PEGylated antisense agent induces RNA degradation through an RNase H mediated mechanism. AA and AA-5K PEG (10 nM) were incubated with a radiolabeled RNA substrate (1 μM ) and RNase H (1 U) for 1 h at 37 °C. Samples were analyzed on a 10% denaturing gel and imaged with a Typhoon FLA 7000. AA: non-modified antisense agent. AA-5K: 5 kDa PEGylated antisense agent.
