Light-activation of gene expression using a PhotoPEGylated antisense agent. HEK 293T cells were transfected with pEGFP-N1, pDsRed-Monomer, and antisense agent. Cell were irradiated for 2 min (25 W, 365 nm) or kept in the dark. A. The cells were imaged after 48 h. The DsRed channel is shown above the EGFP channel. B. The cells were then lysed, and the fluorescence was measured on a BioTek plate reader. DsRed (557/585 nm) fluorescence was normalized to EGFP (485/507 nm) fluorescence. Error bars represent standard deviations from three independent experiments. AA-HP: hairpin antisense agent. AA: non-modified antisense agent.
