Differential conformational changes in promoter selection by mitochondrial RNA polymerase. Schemes show that Rpo41 (blue) by itself binds the promoter (A), mutant promoter (B), and non-promoter (C) with similar Kd values and induces similar DNA bends in all DNAs. No specific structural interactions between the RNAP and DNA are implied by the cartoons. Mtf1 (pink) binding strengthens DNA binding, but only the promoter DNA is sharply bent and melted, whereas the mutant is slightly bent, and the non-promoter remains unbent. The Kd values shown here are based on the equilibrium titration assays.
