The ATPase activity of Isw2 is required for its function in response to replication stress. Strains were grown to saturation, and then 5-fold serial dilutions were plated on YPD plates with or without 50 mM HU, 0.01% (vol/vol) MMS, or 0.02% (vol/vol) MMS. Plates were grown for 2 days at 30°C. The relevant genotypes are shown on the left. Two independent isolates each of isw2 nhp10 and isw2-K215R nhp10 strains are shown.
