Apratoxin S4 (1e) inhibits cotranslational processing in vitro. Products of in vitro translation reactions (rabbit reticulocyte, amino acids, [35S]methionine, canine pancreatic microsomal membranes, mRNA template, and apratoxin 1e) were separated by SDS-PAGE to autoradiographically detect effects on translation and glycosylation of α-factor and signal peptide cleavage of β-lactamase. When PDGFR-β cDNA was used as a template, in vitro transcription/translation was carried out using T7 TNT Quick Master Mix and glycosylation determined by SDS-PAGE followed by autoradiography.
