Reciprocal mutations leading to conversion between monofunctional EPC and bifunctional SM/EPC reactions in TbSLS2 and TbSLS4. A, SDS-PAGE analysis of expression for the different TbSLS variants with mutations indicated. B, TLC analysis of products given by the different TbSLS variants using NBD-C6-CM as substrate. One mutation converts TbSLS2 from an EPC synthase to a bifunctional SM/EPC synthase. Additional mutations inactivate TbSLS2. Two reciprocal mutations convert TbSLS4 from a bifunctional SM/EPC synthase into an EPC synthase.
