Each of these template RNAs possesses a four-base insertion (CTAG) at PV nt 2474 to prevent translation of the P2 and P3 replication proteins. DJB1 possesses two 5′-terminal G residues that inhibit the initiation of positive-strand RNA synthesis (3, 23). DNVR38 and DNVR36 possess authentic 5′ termini. (B) Translation of PV RNAs. Proteins were synthesized in reaction mixtures containing [35S]methionine and DNVR2 RNA (lane 1), DJB1 RNA (lane 2), DNVR38 RNA (lane 3), DNVR36 RNA (lane 4), or no RNA (lane 5). Radiolabeled proteins fractionated by SDS-PAGE were detected by phosphorimaging.
