(A) Hek293T cells were transfected with mCherry alone, mCh-LAPTM4b-WT, mCh-LAPTM4a-WT or mCh-LAPTM5-WT (red). 24 hrs post transfection cells were fixed and stained with anti-Lamp1 antibody (blue) and Phalloidin (green). (B) Additional images illustrate the changes in actin-based membrane protrusions associated with mCh-LAPTM4b-WT expression (relative to mCherry-alone expressing cells) in two independent experiments (1 and 2). All images were taken with LSM510 and analyzed by Volocity 5.4.1 (Perkin Elmer). White arrows indicate examples of actin-based membrane protrusions in which mCh-LAPTM4b is present.
