Maturation of splenic DC following L. donovani injection. C57BL/6 mice were injected (i.v.) with a control solution (PBS) (Untreated), with CpG-ODN (50 μg), with mitogenic anti-CD3 (50 μg), or with 5 × 10⁸ (5 10⁸), 1 × 10⁸, 5 × 10⁷ (5 10⁷), or 1 × 10⁷ L. donovani parasites. At 9 h after treatment, mice were sacrificed and spleens were collected. Pooled low-density spleen cells from three individuals in each group were double stained with PE-labeled anti-CD11c and with biotinylated anti-CD40, anti-CD80, anti-CD86, and anti-MHC class II followed by streptavidin-APC treatment. The data represent the fluorescence of cells gated for high levels of SCD11c expression. These results are representative of four independent experiments.
