Leishmania-induced maturation of DC is not due to internalization of Leishmania parasites by DC. (A) C57BL/6 mice (n = 3) were injected (i.v.) with 10⁸ wild-type L. major or 10⁸ GFP⁻ L. major parasites. At 6 h later, mice were sacrificed and spleen cells were analyzed by cytofluorometry for GFP expression. (B) In the same experiment, spleen cells were stained with the following combinations of antibodies followed by streptavidin-peridinin chlorophyll protein (PerCP) treatment: APC-labeled anti-GR1 and PerCP-labeled anti-CD11b, PE-labeled anti-CD11c and PerCP-labeled anti-CD11b, APC-labeled anti-CD8α and PE-labeled anti-CD3ɛ, and APC-labeled anti-B220 and biotinylated anti-MHC class II.
