X-band EPR spectra of Mn2+ bound to hammerhead ribozymes. Inset shows an overlay of the sixth (highest-field) line of the EPR spectra of Mn2+ bound to WT HHRz, HHRz with a dG10.1 substitution, and Mn2+ in a buffer solution (Standard). Spectra were obtained at 10 K, with 250–350 µM HHRz samples in 1 M NaCl, 5 mM TEA, pH = 7.8, and 20% ethylene glycol. EPR instrument conditions were X-band (9.4 GHz), 0.2 mW microwave power, 100 kHz magnetic field modulation at an amplitude of 15 G.
