The plasmid pETORPHI is an efficient template for DNA synthesis. (A) The assays were done as described in Materials and Methods using the amounts in nanograms of plasmid pETORPHI or Φ29 TP-DNA templates indicated in the figure. The absence or presence of p6 in the reactions is indicated. The size of the amplified bands is shown: 19.2 kb for Φ29 TP-DNA and 5.8 kb for pETORPHI cut with DraI. Lambda phage DNA cut with HindIII is shown as molecular weight marker (Mr). (B) The presence of TP is essential for amplification. The assay was performed as in A, but using p6 in all cases.
