Validation of a subset of genes assigned to the inflammation/stress category. A, PRC dependence of CCCP induction was validated for a subset of seven genes from the inflammation/stress category by measuring their response to CCCP in a second independent lentiviral transductant (PRC shRNA4) exhibiting ∼85% PRC silencing. The three transductant lines (control shRNA, black bars; PRC shRNA1, open bars; PRC shRNA4, gray bars) were treated for 16 h with CCCP as described under “Experimental Procedures,” and the -fold induction for each gene relative to the untreated control was determined by quantitative real time PCR. The insert shows the relative PRC expression in each transductant line. B, HEK293 cells were either untreated or treated with CCCP for 16 h, and PRC protein was assayed by immunoblotting (insert). The CCCP-dependent expression of the subset of seven inflammation/stress genes was compared with that of the respiratory genes described in the legend to Fig. 2 by quantitative real time PCR.
