The oligosaccharide chains of CtmPrP are sensitive to digestion with endoglycosidase H. Detergent lysates of transfected BHK cells expressing wild-type or FLAG-L9R/3AV PrP were incubated without enzyme (lanes 1 and 4), with endoglycosidase H (lanes 2 and 5) or with PNGase F (lanes 3 and 6). Proteins were then precipitated with methanol and analyzed by Western blotting by using antibody 3F4. The band in lane 6 indicated by the asterisk is a proteolytic breakdown product.
