Single-chain proteins and wt-S100A4 exhibit similar Ca binding affinities in the presence of myosin-IIA. Ca2+ binding affinities were measured using a chromophoric calcium chelator 5,5′Br2-BAPTA competition assay. The absorbance at 263 nm was monitored as a function of the Ca2+ concentration: (A) wt-S100A4, (B) sc-S100A4–5, and (C) sc-S100A4–10. The data represent the average from three independent experiments. The insets show the saturation curve representation for the best fit in Caligator.
