Mutation of the BRS motifs in TCRζ alters TCR mobility and its distribution with respect to Lck. (A) Fluorescent recovery trace after photobleaching a 1.3 ×1.3 μm area on the surface of BW5δ cells expressing the indicated TCRζ* construct. An exponential recovery equation was fitted to the data to determine the recovery lifetime and mobile fraction (Materials and Methods). (B) Mean TCR recovery lifetimes from multiple (n = 30 cells) determinations. (C) Confocal imaging of BW5δ cells expressing Lck-DsRed and TCRζ* with or without (B−) the BRS motifs. The images obtained by confocal microscopy represent an optical section of 0.5 μm in z axis, focusing on the glass coverslip. (D) Pearson coefficients representing colocalization of Lck-DsRed with TCRζ* or TCRζ*B−. Data are means of multiple (n = 30 cells) determinations.
