(a) A linear (R2 = 0.94) increase in apparent Km is observed over the range of 0 to 10 μM concentrations of neoamphimedine, indicative of a competitive mode of inhibition; (b) Competitive inhibition studies with ATP depicting a shift in the apparent Km from 390 ± 50 to 790 ± 120 μM ATP. Using the unpaired, two-tailed t-test analysis, this shift in apparent Km was statistically significant (P = 0.010) while the Vmax for both curves was not (P = 0.186); (c) Dose response curve for the inhibition of TopoIIα ATPase function the IC50 is found to be 2 μM.
