Primary mouse keratinocytes, after culturing for five days, were transfected with either non-targeting siRNA (control siRNA) or β-adducin specific siRNA (Addu-siRNA), respectively. The siRNA-transfected keratinocytes at 48 h post-transfection were collected for both western blotting analysis and immunofluorescence labelling. (A). Western blot analysis for expression of spectrin, adducin, P-adducin(Ser662), PKCδ and P-PKCδ(Thr505). Actin was used as a loading control. Results are representative of two separate transfection experiments, with 6 replicates in one transfection experiment. (B).
