IFN-β upregulates tetherin expression in neuroblastoma cells, but has no impact on tetherin shRNA-treated cultures. (A) Tetherin mRNA expression (100 bp) was detected after either medium or IFN-β treatment, of control neuroblastoma cells and tetherin shRNA cultures. Cell lysates were prepared from medium-treated control neuronal cells (lanes 1–3), IFN-β-treated control cells (lanes 4–6), medium-treated tetherin shRNA cells (lanes 7–9), and IFN-β-treated tetherin shRNA cells (lanes 9–12). β-Actin was used as a housekeeping gene (200 bp). All experiments were performed in triplicate and replicated at least three times. (B) Western blot analyses were conducted to study tetherin expression. Cell lysates were prepared from medium-treated control neuronal cells (lanes 1–2), IFN-β-treated control cells (lanes 3 and 4), medium-treated scrambled shRNA-treated neuronal cells (lanes 5 and 6) medium-treated tetherin shRNA cells (lanes 7 and 8) and IFN-β-treated tetherin shRNA cells (lanes 9 and 10). Tetherin was observed at 28 kDa and β-actin at 36 kDa. All experiments were performed in triplicate and replicated at least three times.
