Subcellular localization of Prx IV-S and Prx IV-L. A, a testis homogenate prepared from 20-week-old C57BL/6J mice was subjected to differential centrifugation at 1,000 × g, 10,000 × g, and 105,000 × g. The whole homogenate (Whl) as well as the pellets obtained at each centrifugation step and the final supernatant (Sup) were subjected to immunoblot analysis with antibodies to the COOH-terminal region of Prx IV and to the indicated proteins. B, the pellets obtained after centrifugation at 10,000 × g and 105,000 × g were incubated with various concentrations of proteinase K in the absence or presence of 0.5% Triton X-100 after which the reaction was stopped by the addition of PMSF and the samples were subjected to immunoblot analysis with antibodies to the COOH-terminal region of Prx IV, to Prx IV-L, and to PDI.
