Astrocyte cultures were treated with control or the specific miRNA inhibitors (Applied Biosystems) at 25 nM for 48 h as described in the Materials and Methods, and then stimulated with IL-1/IFNγ. (A, B) miR-155 and miR-155* expression as determined by TaqMan Q-PCR were completely inhibited by specific anti-miRNA inhibitors. (C, D, E) Q-PCR was performed to determine the level of TNFα, IL-6 and IL-8 mRNA expression in the presence of control or specific miRNA inhibitors. Data are expressed as fold changes in expression and then normalized to the levels in control α-miR-treated cultures (Ctr = 1). Both anti-miR-155 and anti-miR-155* significantly inhibited expression of these cytokines. ** p<0.01, *** p <0.001
