(A, B) Photomicrographs to illustrate the aspect of a E17 Shh−/− embryo compared to a E17 wild type embryo. The mutant embryo is not viable due to multiple malformations in particular of the head. Scale bar: A = 1,5 mm; B = 4 mm. (C) The relative level of pMCH mRNA was assessed by quantitative real-time PCR in one Shh+/+ and one Shh −/− E13 embryos. pMCH mRNA level was 85% lower in the mutant. (D) The pMCH mRNA level was also decreased by 90% in E13 embryos two days after injection of the Shh pathway inhibitor cyclopamine (cyclo) to the pregnant mice (injection at E11), as compared to control (injection of DMSO, ctrl).
