Glu-tRNAGln is an inhibitor of the aminoacylation reaction catalyzed by GluRS2. Glu-tRNAGln was formed as described in ‘Materials and Methods’ section and added to the GluRS2 reaction mixture at concentrations varying from 0 to 7 µM. Vi/V0 is equal to 2/[2 + (I/Ki)] when the substrate concentration is equal to its KM (15). Two independent experiments were conducted (filled square, filled circle) with 1 µM uncharged tRNAGln as the substrate, yielding an average Ki of 1.3 ± 0.2 µM.
