Depletion of hSAS6 blocks not only GFP–STIL-induced centriole amplification, but also the recruitment of STIL to the procentriole. U2OS-based GFP–centrin expressing cells (A) or GFP–STIL-inducible cells (B) were transfected with control siRNA (siControl) or siRNAs against hSAS6. Forty-eight hours after transfection, cells were synchronized by aphidicolin treatment with or without tetracycline for another day, and analysed by immunofluorescence confocal microscopy using indicated antibodies (A, B) or by immunoblotting (C). Depletion of hSAS6 perturbs the localization of endogenous STIL to the procentriole. Quantification of centriolar signals of STIL (A, right panel) in siControl and sihSAS6-treated cells.
