DSCR1 (A, from(13)) and PROCR (C) mRNA expression is upregulated in response to PDGF-BB stimulation (30ng/mL). Transcriptional activation, however, is attenuated upon treatment with either CsA (3μM) or A-285222 (10μM). Similar results are seen with use of the VIVIT peptide; DSCR1 and PROCR expression is inhibited at 0.5, 1, 3 hrs (B) and 1, 3 hrs (D) upon treatment, respectively. PDGF-BB induced RAPGEF2 gene activation does not appear to be Cn/NFAT-dependent (E, F). Representative graph shown for A, C, E. Target mRNA levels are normalized to the 18S housekeeping gene.
