(A) NFAT-luciferase activity was measured in control or TG (2 μM)-treated neonatal cardiomyocytes. Increases in transcriptional activity were abrogated by 2-APB (30 μM). (B) RNAi knockdown of STIM1 (Ad-shSTIM1) in NRVMs blunted NFAT activity at both basal and TG-induced conditions. (C) Constitutively active STIM1 (Ca-STIM1) increased NFAT activity, which could be blocked by Cyclosporin A (CSA, 10 μM).
