Gα proteins are not required for Wnt/β-catenin signaling. MEF cells were isolated from mice carrying indicated floxed alleles and infected with either a GFP- or Cre-adenovirus (Ad). Loss of protein was confirmed by Western blot analysis. Stimulation of the Wnt/β-catenin signaling by increasing amounts of Wnt3a conditioned medium (CM) or 40 mM LiCl demonstrates normal Wnt/β-catenin signaling activation in all cases. (A) MEF cells were isolated from sflox/flox mice. (B) MEF cells were isolated from qflox/flox; Gα11KO mice. Cells were pretreated with pertussis toxin (PTX; 100 ng/mL) to inhibit Gαi/o signaling. (C) MEF cells were isolated from 12KO; 13flox/flox mice.
