a) Recombinant PAD4 and PRMT3 were expressed in HEK 293T cells and cell lysates were subjected to a polysome profiling analysis. The profile was performed using a 5–46 % sucrose density gradient and recorded by continuous measurement of absorbance at 254 nm. The positions of 40S, 60S subunits, monosomes (80S) and polysomes are indicated. Fractions were collected, precipitated by tricholroacetic acid and used for Western blots with anti-PAD4 (top panel), anti-PRMT3 (middle panel) and anti-RPS2 (bottom panel) antibodies. b) Polysome profiling of endogenous PAD4, PRMT3 and RPS2 was performed as described in (a).
