Regulation and reversibility of Vps21-mediated liposome tethering. (a) GEF-stimulated tethering. Tethering by GDP-loaded Vps21-decorated liposomes was measured by QLS following the addition of 0.2 mM GTP and varying concentrations of Vps9. Bars span mean + s.e.m.; data points from three independent experiments were binned into 10 min intervals. (b) GAP-mediated reversal of tethering. GTP-loaded Vps21-decorated liposome tethering was measured by QLS following the addition of Gyp1TBC or (*) Gyp1TBC-R343K.
