The parental plasmids pZS21 and pZA31 are described elsewhere (27). Descriptions of plasmids pRBS01 and pBb are available at the registry (http://registry.jbei.org). The copy numbers determined for pBBR1 and p15A ori in pBbB5c and pBbA5a are listed in the registry; they range from 8 to 10 and from 17 to 20 copies per cell, respectively.

aroBop is the codon-optimized variant of aroB. T1 is the E. coli rrnB terminator t1. It precedes Ptrc to prevent run through by the first promoter.
