Knockdown of eIF3a enhances EGF induced c-Fos expression and DNA synthesis. (A) HEK293 cells were transfected with siRNA against eIF3a, serum starved, and treated with 20 ng of EGF/ml for the indicated time points. Induction of c-Fos protein and levels of eIF3a were assessed by Western blotting. β-Actin served as a loading control. The data are representative of three independent experiments. (B) The effects of eIF3a knockdown on DNA synthesis in serum-starved and EGF-treated HEK293 cells was measured by BrdU incorporation. The experiment was performed three times, and >400 cells were counted in each case. The data are means ± the SEM. ∗, P < 0.05. The eIF3a knockdown efficiency was monitored by Western blotting.
