Mild reduction inactivates the ability of 19-kDa GP pseudovirions to mediate infection. VSV GPΔ pseudovirions and VSV GPΔ pseudovirions that had been cleaved with thermolysin to generate 19-kDa GP (VSV-19kDa GP) were pretreated at pH 7.0 or at pH 5.5 with or without 5 mM TCEP for 10 min at 37°C. Samples were neutralized by adding 16 volumes of Optimem medium and added to Vero cells. After 24 h, the cells were detached and fixed, and the percentage of GFP-expressing infected cells was measured by flow cytometry. Data are from one of two experiments (performed in duplicate) with similar results. Error bars indicate SD.
