Whole-cell protein extracts from HEp-2 cells analyzed by immunoblotting showed that (A) IFNγ mediated (50ng/ml, 0.5h) Stat-1 tyrosine phosphorylation is suppressed by enterohemorrhagic Escherichia coli O157:H7, strain EDL933 (EHEC), but not by enteropathogenic Escherichia coli O127:H6, strain E2348/69 (EPEC) (MOI of 100∶1; 6h). (B) Incubation with sterile culture supernatants (6h) from EHEC, but not EPEC, suppressed IFNγ-mediated tyrosine phosphorylation of Stat-1. (C) Incubation with chloramphenicol (100 µg/ml) up to 1h after infection with EHEC (MOI 100∶1; 6h) prevented EHEC suppression of IFNγ mediated Stat-1 tyrosine phosphorylation.
