CaSR-mediated β-catenin Ser-552 phosphorylation and plasma membrane localization. NCMiCaR cells incubated without (−) or with (+) doxycycline (0.1 μg/ml) were challenged with 5.0 mm Ca²⁺ or 100 nm R-568 in a background of 1.4 mm Ca²⁺ for 1 h and processed for immunofluorescence using a rabbit Ser(P)-552 antibody and mouse monoclonal antibody anti-total β-catenin. The images displayed are representative of 90% of the population of cells. Scale bars, 10 μm. Quantification was performed as described under “Experimental Procedures.” The values represent the MI ± S.E., and they were compared using unpaired Student's t test (**, p < 0.005; *, p < 0.036).
