Overexpression of miR-145 inhibited VEGF and HIF-1α expression. (A) SW1116 and SW480 cells overexpressing miR-145 were analyzed for VEGF mRNA levels by TaqMan real-time PCR. Results are presented as mean ± SE of VEGF mRNA levels from three duplicates. Asterisk indicates significant difference when compared to the control (P < 0.05). (B) Mock cells, cells transfected with pLe-SC and pLe-miR-145 were used to test HIF-1α mRNA levels by semi-quantitative RT-PCR. (C) Total cellular proteins were collected and subjected to western blotting. Forced expression of miR-145 increased PDCD4 level and inhibited HIF-1α and VEGF protein expression. (D) The protein levels of above study from three independent experiments are quantified and presented as mean ± SE. Asterisk indicates significant difference when compared to the control (P < 0.05).
