Induction of cell death in a mixed population of cells promotes enrichment of genome-positive cells. (A) CD34⁺ cells (3 × 10⁵) mock- (M) or HCMV (V)-infected were further incubated with cisplatin (+) or DMSO (−) for 3 hpi. After a further 4 h, cells were plated in fresh media and cultured for 72 h. Viability was assayed by trypan blue. Error bars represent a triplicate counts taken from three independent experiments. (B) DNA was harvested from cultures at 3, 24, and 72 hpi and, following IE and GAPDH PCR, bands were analyzed by densitometry to quantify viral and cellular DNA content.
