Point mutations (Q98L, Q560A, and D568A) were introduced into KIF1Bβ by using the QuickChange XL site-directed mutagenesis kit (Stratagene) following the manufacturer's recommendations. B, colony formation assay. SH-SY5Y cells were transfected with an empty vector or with the indicated expression vectors. Forty eight hours after transfection, cells were transferred into fresh medium containing 500 μg/ml of G418. Two weeks after selection, G418-resistant colonies were fixed and stained with Giemsa solution, and number of drug-resistant colonies was scored.
