Enhanced NBD-PS flip in the dnf1Δdnf2Δ strain is down-regulated by ethanol. Early log phase DNF1DNF2 (BY4141) and dnf1Δdnf2Δ (HCY13) cells (A₆₀₀ = 0. 2-0.4), grown in YPD were resuspended in SDC media with varying concentrations of ethanol. Cells were pretreated with ethanol for 10 min at 30 °C, and then labeled with 5 μm NBD-PC or 10 μm NBD-PS at 2 °C for 1 h. Cells were washed three times with 2 volumes of SC-azide and analyzed by flow cytometry. Data presented are the mean of at least three independent experiments, and the error bars represent the S.D. between experiments.
