In vivo tumor growth of diffuse-type gastric carcinoma cells is accelerated by the expression of dnALK3. A: Diffuse-type gastric carcinoma cells were infected with lentivirus carrying GFP cDNA (OCUM-12-GFP and HSC-39-GFP) or HA-tagged dnALK3cDNA (OCUM-12-dnALK3 and HSC-39-dnALK3). Parental cells and infected cells were treated with BMP-4 for 1 hour. Cell lysates were subjected to immunoblotting with antibodies, as indicated under Materials and Methods. B: Diffuse-type gastric carcinoma cells expressing GFP or dnALK3 were treated with TGF-β1 for 1 hour. Cell lysates were subjected to immunoblotting with antibodies, as indicated under Materials and Methods. C: Diffuse-type gastric carcinoma cells expressing GFP or dnALK3 were treated with BMP-4 for 24 hours. Expression of ID3 mRNA was determined by quantitative real-time RT-PCR. Data are presented as means ± SD. D: Diffuse-type gastric carcinoma cells expressing GFP or dnALK3 were subcutaneously xenografted into BALB/c nu/nu mice (OCUM-12-GFP, n = 7; OCUM-12-dnALK3, n = 9; HSC-39-GFP, n = 7; HSC-39-dnALK3, n = 8). Quantification of tumor volume over time (OCUM-12, day 0 to day 42; HSC-39, day 0 to day 19) are shown. Data are presented as means ± SE. *P < 0.05; ***P < 0.001.
