Sh3rf2, unlike POSH, binds to both active and inactive Rac1. POSH and Sh3rf2 were transcribed and translated in vitro in the presence of radiolabeled methionine. An aliquot was reserved as input. Products were incubated with beads bound to either NRac-GST or VRac-GST fusion proteins or uncoupled GST, washed, and resolved by SDS-PAGE and visualized by autoradiography. Smaller bands represent incompletely translated protein, which is still capable of interacting.
