Pan-SHIP inhibitors reduce viable cell numbers of SHIP2-expressing breast cancer cells. MDA-MB-231 (A) and MCF-7 (B) cells were treated with indicated concentrations of pan-SHIP inhibitors for 2 h. Cells were washed twice, and fresh medium was added. Viable cell number was determined with a cell counting kit after 24 h, and results represent the mean of at least two independent experiments. —●—, 3AC; Inline graphic, 1PIE; Inline graphic, 6PTQ; —▼—, 2PIQ. (C–E) After treatment of MCF-7 cells with 10 μmol/L 1PIE, 2PIQ or 6PTQ for 2 h, cells were subsequently cultured for 36 h in the absence or presence of the indicated concentrations of PtsIns(3,4)P2 or PtdIns(3,5)P2, after which cell viability was determined with a cell counting kit. The mean of three independent experiments is shown. Inline graphic, μmol/L 3AC for 2 h, after PI(3,4)P2; —■—, PI(3,5)P2. (F) MCF-7 cells were treated with 10 which the medium was replaced and cells were cultured in the absence or presence of indicated concentrations of PtsIns(3,4)P2 or PtdIns(3,5)P2 for 36 h. Cell viability was determined with a cell counting kit. Inline graphic, PI(3,4)P2; —■—, PI(3,5)P2.
